NAC and Glutathione: Supporting Your Body’s Primary Antioxidant System
Most antioxidant supplements try to mop up free radicals from the outside. A more useful approach is helping your cells make more of their own primary antioxidant, glutathione. NAC is one of the most practical ways to do that.
What NAC Is
NAC stands for N-acetylcysteine. It is a stable form of the amino acid cysteine. Your body uses cysteine as a key building block for glutathione, the central antioxidant that works inside cells. NAC has been used in medicine for decades, but the reason it matters for everyday health is its ability to raise glutathione levels. It is inexpensive and easy to take as an ordinary oral supplement.
How It Works
Glutathione is made inside your cells from three amino acids: glutamate, cysteine, and glycine. Of these three, cysteine is usually the one that runs short first.
NAC supplies extra cysteine in a form the body can absorb and use. Once that cysteine is available, cells can produce more glutathione. Higher glutathione improves the cell’s ability to neutralize harmful reactive oxygen molecules, keep mitochondria working cleanly, support natural detoxification processes, and help regulate inflammation signals.
NAC also has some direct antioxidant effects of its own and can influence how cells handle glutamate. The main benefit comes from boosting the body’s own glutathione production.
Making It Work Better
NAC covers the cysteine step, but the glutathione system works more effectively when a couple of other pieces are in place.
Glycine is one of them. Glutathione needs glycine as well as cysteine. In older adults and during periods of higher stress, glycine can also become limited. Taking glycine together with NAC (often called GlyNAC) raises glutathione more effectively than NAC alone.
The strongest human data comes from trials at Baylor College of Medicine. In older adults who started with low glutathione, 16 to 24 weeks of GlyNAC roughly doubled glutathione levels, sharply lowered oxidative stress markers, improved mitochondrial function, reduced inflammation, and led to better walking speed, strength, and some cognitive measures. When people stopped taking it, many of the benefits faded. A larger short-term study in healthier older adults found clearer benefits mainly in those who already had higher oxidative stress and lower glutathione at baseline. Safety was good across these trials. The research doses were higher than typical single capsules, several grams of each amino acid per day in the weight-based studies.
Selenium is another useful partner. It is required for the enzymes that actually use glutathione to break down hydrogen peroxide and other damaging molecules. Without enough selenium, the extra glutathione you make is less useful. This is one reason selenium status matters for overall antioxidant capacity, including in tissues under higher oxidative demand such as the thyroid.
Other helpful supports include alpha-lipoic acid, which helps recycle glutathione, and simply eating enough protein so the body has a steady supply of amino acids.
What the Benefits Look Like
The most consistent findings are rises in glutathione itself and improvements in overall antioxidant capacity. Oxidative stress markers (such as lipid peroxidation products) tend to fall when glutathione levels go up.
In the GlyNAC trials with older adults, these biochemical changes came with clearer practical effects. Mitochondrial function improved. Inflammation markers dropped. People walked faster, showed better muscle strength, and performed better on some cognitive tests. Body composition and exercise capacity also moved in a positive direction. Many of these gains faded when supplementation stopped, which suggests the effects depend on continued support of the glutathione system.
Beyond the aging trials, NAC has a longer track record of supporting lung and airway health by helping thin mucus and reducing oxidative burden in the respiratory tract. It also appears useful for recovery from physical stress, where oxidative load and inflammation are elevated.
When inflammation or oxidative stress runs high, including in some autoimmune and thyroid-related situations, supporting glutathione production is a rational approach. The evidence here is still developing, but the underlying biology lines up.
Practical Notes and Dosage
A typical supplemental range for NAC alone is 600 to 1,800 mg per day, often split into two doses. Most people use the ordinary oral form. For the GlyNAC approach, the research used higher combined amounts of glycine and NAC. Selenium is usually kept in the 100 to 200 mcg supplemental range. Taking NAC with food can improve tolerability for some people.
NAC is generally well-tolerated. The most common side effects are mild digestive upset. Taking NAC is a straightforward way to support your cells’ own glutathione production. Pairing it with glycine and adequate selenium addresses more of the pathway. The real advantage of NAC is working with the body’s built-in antioxidant system rather than relying only on external scavengers like supplemental vitamins and even supplemental glutathione.
